It can be any funny scene, funny line of dialogue or simply funny reactions from the characters (of which there have been many over the years).
My top moment would have to be this one (though I’m not sure what episode/season it’s from, since everything after Season 5 pretty much blurs together in my mind).
Some other funny moments which immediately spring to mind...
This Sam moment from Season 2’s ‘Tall Tales’.
This scene from Season 3’s ‘Mystery Spot’ (I wonder how many takes this one took for them)...
And lastly, Sam’s/Jared Padalecki’s reactions in this scene.
