Is it possible that Will Ferrell and John C. Reilly are now #1?
No?
Then - where go you?
Go!

I would say that Reilly and Ferrell are the top one these days. Glad to see them doing another movie together, even though it's impossible to top Step Brothers.
The best from 30 years ago? How bout Belushi/Aykroyd or Pryor/Wilder? I wish Steve Martin and John Candy had done more together, they were so great in Planes, Trains and Automobiles.
The best all-time? Abbott/Costello or Laurel/Hardy, though the greatest comedy team was easily The Marx Brothers.

amc had Planes, Trains and Automobiles on a week after Thanksgiving.
I don't know if there's any legal stuff involved here - i.e. a channel can't get the rights - but this movie deserves a 24/7 nonstop play on Thanksgiving like Groundhog Day with Bill Murray.
Martin and Candy play off each other tremendously.
You mentioned Abbott/Costello.
I'm a horror fan. Ever see Abbott and Costello Meet Frankenstein?
Tremendous - pretty much a Monster Mash with Dracula, the Wolf Man an the Invisible Man too.............

Of course I've seen Abbott and Costello Meet Frankenstein, it's a classic. Of course, I just realized I forgot to mention Cheech and Chong? That's cuz I haven't gotten stoned yet today. Time to change that, we've got Sixers/Raptors in an hour......

and i'm thinking outside the box here - but they play off each other tremendously - which can in turn create comedy cold.
haters will hate -
I always loved Damon and Affleck.
this is one of the greatest scenes in cinema history IMO:

If the script was right, I'm sure those two could do a fantastic comedy together. How bout Will Ferrell and Mark Wahlberg? They were great in The Other Guys and seem to work well in those family comedies they've done together.

In the commercial for the new Reilly/Ferrell movie, they are actually showing clips from Talladega Nights and Step Brothers......that doesn't really say much about this new movie, tbh.

Laurel and Hardy by a country mile.
Second? I'm very fond of the Two Ronnies.

Simon Pegg and Nick Frost for the current guys in movies. Although without Edgar Wright they really haven't done anything of note.
Mitchell and Webb, pretty excellent, but I don't know that they've done movies together.
Jack Black and Kyle Gass for music comedy duo.
Trey Parker and Matt Stone for writing/animating/Broadway musical creating.
But for the classic on the silver screen old fashioned comedy duo it's Laurel and Hardy. They played off of each other better than anyone else. They each treated the other as the fool and themselves as the straight man, which is still sort of a novel approach.
