After he reads his mother's file he concludes that the condition he thought he had was all bull made up by his mother, and that the laugh is just part of the real him.
Is this true though? It would mean that all of the moments before were times when he genuinely found the situation funny and was trying to suppress genuine laughter. Was he genuinely laughing during the subway scene for instance? It seems weird, but would fit the character of Joker from the comics quite well.
Or is the truth somewhere in between? What say you?
Its hard to tell. Like, how about that scene at work when the big dude is making fun of the little dude and Joker laughs louder than anyone, but shuts it off immediately after turning the corner. I wonder...

I haven't seen the film yet but from what I heard it's not that he doesn't have a condition just that the cause of his condition isn't what he was told, like he believes it's a natural issue he's been born with, which turns out to be a neurological issue that came about due to childhood abuse by his mother.
Though as I said that's just by piecing bits together, I could easily be wrong.

And his laugh is a lot more natural and less painful after he finds out, almost as though he has now learned to let the real version of himself out, where as before he was suppressing it. Further evidenced by the fact that he now laughs at horrible things, implying that he genuinely finds them funny
