I would like to know what really happened to Amy Dudley (née Robsart) or who killed the princes in the tower.

It's got to be JFK - not because of the 'magical bullet' or 'the grassy knoll', but because of the killing of Lee Harvey Oswald by a guy with links to organised crime. There is no satisfactory explanation for that.

There's a copycat killer in relation to this. Stella Nickell was some broad who poisoned her husband, and to blame it on Tylenol she went around to random pharmacies poisoning their supplies of the pills. In short she killed other random innocent people to cover herself up. I think that's the story. Anyway, evil!
Wiki- Excedrin capsules in Washington state were tampered with, resulting in the deaths of Susan Snow and Bruce Nickell from cyanide poisoning and the eventual arrest and conviction of Nickell's wife, Stella, for her intentional actions in the crimes and connected to both murders.

Another murder that I would like to see solved is that of Lynn Harper.

Bradford Bishop, a career State Department officer with a background in military intelligence, murdered his family then disappeared. On two occasions, reliable witnesses claim to have spotted him in Europe.

Msdemos, I voted "other." Forgive me for coloring-outside-the-lines, as my answer isn't exactly to the point of your question. It's not one murder ... it's several.
I live in "ZODIAC" country. The early 70's was a bizarre time, around here. The only murder for which Zodiac claimed responsibility in San Francisco, took place only a couple of miles from where I live now, and that site is on one of my morning walking routes. My Aunt and Uncle lived closer than that to one of the other murder venues.
There are a host of suspects, who have been on that list for years. From time to time, someone comes forward and adds a brother, father, step-father, cousin, etc., to the list. They all seem to have a number of factors/traits that would fit the profile. But, there's never been a definite ID.
