This Something Mary scene was the first that came to mind when i saw the subject title. Re The actual clip shown from o.p..... each to their own and all that, but in all honesty, I am mystified why that is thought of as remotely funny......let alone funniest. Perhaps if I'd smoked a mattress full of good weed........but then i would find even Piers Morgan and Kanye hysterically funny.

Funny scene, but funniest is all subjective. I laughed more when Miller was in the phone booth and the homeless person was yelling at her.... "Get out of my house!"HAHAHA! I forgot about that. I need to rewatch this. Everything at the bus station is gold.

Among the more "random" yet hilarious scenes that I remember seeing, has to be right near the end of the romantic 80s comedy Mannequin (1987), where the leading man is obsessed with a mannequin doll (Kim Cattrall), and yeah, surely things move fast in the magic world of cinema as the doll turns into a stunning real life woman and the couple lives happy for the rest of their lives.
Anyway, the scene that I am very sure took place (not seen it though, for at least a decade or more) is right near the ending, as we see some random worker or janitor, who witnessed this fairytale come true, and realizes that maybe his dream woman is not out there in the crowded streets, but might just be lying somewhere deep beneath tons of garbage. So what does he do? He dives head first into some dumpster in search of his true love, the end.

The entire rock n' roll star trashing done by Belial inside some cheap and sleazy motel in Basket Case (1982), is still one of the funniest scenes I have ever witnessed.
Pure movie magic:
I think Frank Henenlotter was at first embarassed of this scene and special effects, and had no intention of having the audience laugh at this scene and film, but I am glad he decided to reconsider and threw it in, as yeah, it might not be a straight up comedy the entire time, but there is so much ridiculous scenes taking place, so if he was to get rid of every funny part, character or quote, the film would not last for more than a few minutes.

The whole dentist scene from The Pink Panther Strikes Again (1976), where Clouseau visits his former boss and now complete mad as hell Dreyfuss, who suffers from some bad toothache, and in return while heavily drugged, Clouseau pulls out the wrong tooth, and as the laughing gas seems to operate on not just the patient but also the practioner, and where his face suddenly stars meltin and yeah, no matter how many revisits, it is still surely among the very top of funny and classic comedy movie scenes:
