Generally, no. There are a couple of movies that I've watched over the years that I've found difficult to understand and they proved to be frustrating viewing experiences. Some examples would be Memento (2000), Cloud Atlas (2012) (the ultimate example) and even 2001: A Space Odyssey (though I did like the latter film).
An exception would be Dario Argento's Suspiria (1977). I didn't entirely understand everything that was happening due to its dreamlike nature, but thoroughly enjoyed the experience nonetheless.
