Julianne Moore in THE HAND THAT ROCKS THE CRADLE I thought, "Who does this actress think she is?" Now I'm madly in love with her.

All seen first run in the theater:
Herbert Lom in Mysterious Island.
Sean Connery in Dr. No; and character actor Joseph Wiseman as the eponymous "Doc".
Robert Shaw in Battle Of The Bulge.
Michael Caine in The Ipcress File
Samantha Eggar in Return From The Ashes
Lynn Redgrave in Georgy Girl (I believe this was her feature film debut).

Harvey Keitel in "Saturn 3." I had heard his name a few times but this was the first movie I saw him in. A little afterward I saw him in "The Border" and "The Duelists."

I probably saw Tomas Milian first in JFK, but I didn't notice him until THE BIG GUNDOWN, and yet, he had one of the most important roles in THE AGONY AND THE ECSTASY as Raphael (and some important lines too) which I had seen a few times before The Big Gundown.
On a completely different track, I first noticed Famke Janssen as an unnamed model in a magazine back in the late 80s and thought--she could be in movies. A few years later, she was.

I first saw Bob & Carol & Ted & Alice (1969) as a midnight sneak preview, in Denver, with a group of other young singles. We went to see the main feature--I can't remember what it was now--and stayed to watch B&CT&A on a Saturday night. We all loved the comedy of the movie. Of course we knew Natalie Wood from her movies and Robert Culp from his TV show I Spy. But, who were these other two actor that made us laugh so hard: Elliott Gould (oh he is Barbra Streisand's husband) and Dyan Cannon. Both actors won us over immediately.
I think these two actor's careers were launched from this movie. Elliott Gould had a leading role in MASH (1970). Dyan Cannon only had to wait a little longer.

I first time I saw Joan Blondell was in "Grease" (1978)... But didn't "care"... The next film is saw Joan Blondell in, was "Gold Diggers Of 1933" (1933). I wanted to see that one was because Ginger Rogers was in it... But this time Joan Blondell caught my eye, and I became a fan! Ginger Rogers & Joan Blondell also starred together in "Broadway Bad" (1933)... but I haven't seen that one yet (Not On DVD)... Not Long Ago, I purchased a Four Movie/Four Disc DVD Set called "Busby Berkely Musicals"... It was the best of both worlds... "42nd Street" (1933) with Ginger Rogers "Footlight Parade" (1933) with Joan Blondell "Dames" (1934) with Joan Blondell "Gold Diggers Of 1937" (1936) with Joan Blondell I can't say I really enjoyed those "Busby Berkely Musicals" movies, but I really didn't buy them for the stories... LOL! Joan Blondell also starred on the "Twilight Zone" episode: "What's in the Box" (1964)

MCDemuth , It's fun finding a less than young actor who has been around for a while and working back through their early days ! Funny you should say that... The first time I saw Rhonda Fleming was in the "The Love Boat" Season 2 episode segment: "Mona Of The Movies" (1978)! Yeah, I know.... NOT A MOVIE... Sorry OP! I loved her performance, and now, I have started watching some of her earlier MOVIES when she acquired the nickname, the "Queen of Technicolor", and I have been enjoying her other performances too! Ms. Fleming is still with us, and she just celebrated her 95th Birthday on August 10th!
