Every second of The Dark Knight. But to narrow it down: him laughing while he falls to his death, him also laughing as Batnan pounds him in the face, his neck crack when he's about to rastle with the cop, him poking his head out of the cop car, the Daffy Duck moment with the detonator, the way he says "I just want her to know that I don't care about the scars!!!", and of course the pencil trick.
Hamill's entrance in Mask of the Phantasm.
The whole Tim Drake flashback in Return of the Joker.

A lot of great ones have already been mentioned. I'll add this one from "Mask of the Phantasm" (1993). I especially like how the Joker just starts laughing in face of what looks like certain death.

I think that that is the Number 1 Most EPIC Joker Laugh of All-Time, Mark Hamill absolutely nailed it for that scene.
