I wouldn't make one. Predator was done right back in 1987.

If I understand the Predator (and I may well not), I'd think an elite hunter, rather than the young one we saw in the jungle, would be able to engage human 'hunters' in the desert. It'd be tough without the cover of the jungle, but an expert might be pretty interesting to watch in the desert. Especially since so many soldiers and tech are being deployed in that area.
Desert Predator would have to take stealth to the next level...due to lack of cover and the advancements in our human military technology.
It'd be interesting to see the Predator in the middle-east. Not a member of religious factions, but just the landscape...and that there is a strong military presence there. Seems like he'd have to hunt in a different, more long-game sort of way. Of course there is civilization and buildings all over the place there, but no trees, a strong human military presence, fewer places to hide if you step in a puddle. Could be cool...
I do think the Predator needs a bit of time on the bench. Everything since AVP has been disappointing to this character. I could be misreading it, but the mystery is one of the most alluring things about the Predator. If we know too much, it isn't as thrilling. If AVP:R did one thing right, it was showing just a glimpse of where the Predator may have come from...

I actually always liked the idea and plot of "PREDATORS" where a bunch of Humans are dropped on a Planet and then Hunted by the Predators.
You could do so many things with that kind of a story.

Predator has to win back his kids in a custody battle.
Predator gets AIDS and is unjustly fired from his law firm. He hires a still xenophobic lawyer to represent him.
Predator is an unstable taxi driver who wishes a real rain would come and wash all the scum away.

Predator's on a hunt to kill POTUS as the ultimate price.
Maybe with a post credit showing them going on a time journey to hunt Hitler.

I wouldn't make one. Predator was done right back in 1987. Yeah, though a backstory film to that one wouldn't be bad. All the dead CIA & Soviet plants in the jungle.

I'm not sure what my ideal sequel would be about, but it would preferably have the following aspects:
1.) Likeable characters. One big reason the original film works and the most recent installment doesn't is due to the characters. I'm not saying Arnold, Carl Weathers, Jesse Ventura and the gang gave Oscar-worthy performances, but they had a macho camaraderie and colorful personalities that made me care about them. Give us some human protagonists we can cheer on and it makes the story more engaging.
2.) Remote setting. The jungle setting in the first film added a sense of dread due to how isolated the characters were in the vast foliage and not knowing where their attacker was. Sure, while the city setting and game preserves in the next two films made for some fun sequences, they never recaptured the same isolated feel of the original (it also didn't help that Predators just remade the first). My ideal sequel would be set out in a vast desert, outpost in the ocean or up in the mountains.
3.) Making the Predator scary again.

The original Predator sequel idea which eventually became PREDATORS was that those aliens which survived an encounter with an alien would be whisked off to the predator homeworld and subjected to a fight to the death for the amusement of a predator audience. Arnie was supposed to be in it.
That was the most promising sequel idea--although they could have done a better Alien vs Predator movie as well. I'd much prefer if they had met for the first time--having the predators using aliens for hunting practice was taken from the late 80s comic--along with the LAME good predator who fights alongside the human.
The original predator was a sore loser. The 1990 sequel made them have a "code."
They painted themselves into the corner by having the predators leave Danny Glover alone and just fly off.

A Predator lands on a planet where it becomes the hunted. Somewhere along the way, it learns valuable life lessons and regains a Predator version of its humanity.

Human's discover the Yautja homeworld and abduct several of them. All of them are killed in experiment's, except for one who manages to escape. The rest of the movie follows it as it crafts weapons out of objects it finds and trys to survive as the police and military hunt it.

A couples of ideas:
Predator movie that takes place during the Vietnam War. US soldiers see a bunch of skinned comrades, assume it's the Viet Cong, slowly but surely they realize it's something else.
Predator movie that takes place during the Wild West. Kinda the same idea as before, cowboys see a bunch of skinned bodies, assume its Native Americans, after a while they realize it's something else. This could be interesting as I'm not even sure the concept of "space aliens" was even that common back then, so the locals aren't sure what they're dealing with, even going as far as thinking the Predator is a demon or some sort of supernatural being.
