This is a multi part question, and you have to consider each character individually. Would Luke Cage ever take a lead role in the Avengers like he does in the comics, and would they need to recast a more popular actor to do so? Could Daredevil ever join Spiderman and would they need an A lister on the big screen to make that happen? Could they try a Punisher movie or team up film and is John Bernthal silver screen material?

There is no need to recast any of the Defenders. I had misgivings about Colter because I was looking for someone more muscular, physically imposing. But he's actually okay as Luke. If there's one Defender who might be recast it is Iron Fist due to the whitewashing BS, but I don't see the need for it.
But I can't see any of the Defenders making it to the big screen.
I don't think there will be another Punisher film for the foreseeable future. We have had three already, and the character's tone is more suited for Netflix than the MCU. Same with JJ. If iron Fist was a hit with critics, a Heroes for Hire might have been considered. Now I think that's more likely on Netflix.
DD might have the best shot, and him teaming up with Spidey sounds cool, but it's a long shot IMO.
Anthony Mackie even said they're different companies, different universes. That's not true but it's an indication of this seeming divide between the movie and TV departments. Even Chloe Bennet feels the same, saying the MCU ignores Shield. I am still pissed AoS didn't have a cameo on AoU when Fury showed up on the Helicarrier. But if the Defenders do appear in the MCU, I don't want any recasting.

The major problems are twofold:
1) Scheduling/Location. The Netflix shows are made in NYC and other parts of the East Coast while the MCU movies are usually made in California or England. It's hard to work the filming all together so everyone is available.
2) Ike Perlmutter may be out of the way, but the head of the TV Department (it's Jeph Loeb, right?) doesn't seem to like Kevin Feige anymore than Perlmutter did.

I like Charlie Cox as Daredevil, shame he'll probably won't appear in the films.

I think all of them stand a chance except for Finn Jones as Danny Rand since almost no one likes his performance.

I was really more referring to the question of recasting, and not the likelihood of these characters actually getting a film.
Would they recast?

I think Charlie Cox has mentioned that he thinks the roles Will be recast if they need version of those characters in the movies

Its said in Cox's contract that if they need him for a film he'll be requested to appear as Matt Murdock, even if its a small role. Its a bit of another story in regards to Daredevil where Murdock has the whole suit on - in that case they don't necessarily need Cox to be there but rather a stunt person, he could audio dubbing for lines if there are any. The contracts are pretty complicated, mostly due to Perlmutter.

They already have Dr Strange & Hulk so all they need is Namor to make the original team. They even have Valkyrie I dug the run where they were called 'The New Defenders.' Great run, it's probably my second favorite title of all time. Such a funky lineup in those days. The stories were pretty good and they were all over the place, fitting for a team that wasn't really a team. Also some of the best comic covers ever during that run.
