A lot of TV stars were made for fame on the little screen (e.g., Farrah Fawcett Majors) and did not have as effective presence when they showed up on the big screen. But a few did, I think.
My pick is Bea Arthur, who was huge in the personality department and was not diminished on the big screen.

I'd say Patrick McGoohan was charismatic enough to be a major film star, but although he did a number of pre-Secret Agent films in the UK, and several more after his SA and Prisoner television stardom, his film career never really took off. It may have been that, handsome and commanding as his screen demeanor was, there was just something a bit too unique and eccentric about his personality to quite 'click' on the big screen.
