Seen this on another site.
At times, a very good character on a TV show wasn't there from the beginning. I'm excluding characters just brought in for one season or a single story arc. Think Phil Leotardo from The Sopranos or Frank Sobotka from The Wire. A few examples...

Norman Buntz from Hill Street Blues
Ben Linus for Lost (it was good then)
Gustavo Fring and Saul Goodman from Breaking Bad

Angel - Fred (introduced at the end of Season 2, made a regular in Season 3).
Star Trek: Voyager - Seven of Nine (introduced/added as a regular in Season 4).

Ensign Ro Laren in ST-The NG. Ro Laren has placed in several polls featuring the characters of the franchise over the course of several years. In a list of the strongest female characters on the official Star Trek website by Jordan Hoffman in 2012, Ro Laren was placed second behind Edith Keeler from the Star Trek: The Original Series episode "The City on the Edge of Forever". He said that Ro was one of the "richest figures in Trek", and wished to be able to see an alternative timeline where Forbes had accepted the main cast position on Deep Space Nine.[46] A poll at the Creation Entertainment official Star Trek convention in 2013, conducted by Hoffman, placed Ro Laren as the fifth most popular recurring character among fans, behind Elim Garak, Q, Gul Dukat and General Martok.

Newman in SEINFELD immediately comes to mind. He was always listed as a guest actor, but he was essentially the 5th main cast member. Also Desmond Hume and Daniel Faraday.

John J. Sheridan was played by Bruce Boxleitner starting in the second season. Marcus Cole was played by Jason Carter in the third and fourth seasons of the show. Zack Allan was played by Jeff Conaway. He regularly appeared in the show from season 2 onwards.
