I say other than Matt Dillon, because I think he'd be the overwhelming first choice.
This is a hard choice among several good characters. as Gunsmoke really seemed to bring out the best in those who portrayed characters on the program. Well, good scripts and good directors would do this.
I'd have to say that my personal favorite by a slight margin would be Quint Asper, as performed by Burt Reynolds. I find him to be engaging and multi-faceted, but without some of the silly aspects of other characters that Marshall Dillon relied upon for help. He basically stood as an equal to the Marshall, and Burt Reynolds always did an admirable job in his portrayal.

My most favorite Gun Smoke character is in the episode named The Returning which is from season 12. My most favorite late singer, Steve Sanders of the Oak Ridge Boys that is, got to be in that episode and it was filmed the year of his 15th birthday which should give you an idea of what character he gave us, one who made a huge difference near the end of the episode. I like Festus Hagan as well, but chose Quint by a small margin.
I prefer the Festus who appeared in the B&W seasons most. Later on in the color years he tended to become less of his own character and more like Chester. And I got more annoyed with each later season as they built up his quarreling with Doc to the point of excess, closing many episodes with their diatribes.
I was disappointed that they would not pay Ken Curtis the amount he wanted to bring him aboard the TV Movie, Gunsmoke: Return to Dodge. And he was not in any of the others, though he did appear in one episode of the How the West was Won TV series with James Arness.

A favorite regular on Gunsmoke: Glenn Strange played bartender Sam Noonan for twelve seasons - 247 episodes - following decades of playing assorted cowboys, villains and henchmen as well as the Frankenstein Monster. He was 6' 5" and a real Texas cowboy and singer.

A favorite regular on Gunsmoke: Glenn Strange played bartender Sam Noonan for twelve seasons - 247 episodes - following decades of playing assorted cowboys, villains and henchmen as well as the Frankenstein Monster. He was 6' 5" and a real Texas cowboy and singer. I liked this character as well. I think he was under-utilized for his size. Often he'd get knocked out or if he grabbed his shotgun, he was already covered or Kitty would tell him to put it away. She was always the wet blanket when this happened. I would have like to seen him actually punch out some rowdy barfly on occasion, but I guess that was always left to the Marshall to do that.

He was 6' 5" and a real Texas cowboy and singer. Glenn Strange's Sam the bartender always had a twinkle in his eye. There was an of innate kindness to him that seemed to come as much from the actor as the character he played. He was popular with viewers too. Odd that he first gained a measure of fame playing monsters in horror movies in the 40s, notably Frankenstein's monster. His background in films was similar to Lon Chaney, Jr.'s, though Strange was never a star attraction the way Chaney was. That Strange and Gunsmoke star James Arness,--both very tell men--played monsters in films--Arness, just once, I believe--causes me to suspect that Glenn Stranger was chosen by Arness for his role in the bartender at the Long Branch.
