You know I must say OF MICE AND ME for this one.  For a newer film, STAND BY ME = boyhood friendships ( I know some don't agree with this one but, whut tha hey !) May seem off the wall at first glance perhaps but, BAMBI. It really is all about friendships. Ditto. THE WIZARD OF OZ. Sticking together in the face of adversity, ie friendship. TOY STORY and follow-ups. More friends!

The first to come to mind, is the excellent Hungarian film from Miklós Jancsó, Így jöttem , My Way Home (1965). In the final days of World War II, a young Hungarian man is making his way home, through a countryside ravaged from war, he is captured and imprisoned by the Russians. and then left in the custody of a young Russian soldier who is tending cows for a dairy farm. The two youths strike up an unlikely friendship and form a close bond in spite of not speaking each other's language Jancsó's portrait of human warmth, the developing relationship between the two men is one of the most moving and clear-sighted analyses of male sensibilities and friendship in all cinema.

Little Caesar / Mervyn LeRoy (1931). Rico’s rise is helped by his friend, Joe and his fall comes because of his breakdown when Joe goes off with a woman.
Angels With Dirty Faces / Michael Curtiz (1938). The story turns on the childhood bond between the adults played by Pat O’Brien and James Cagney – one of whom couldn’t run as fast as the other.
Double Indemnity / Billy Wilder (1944). The friendship between Neff and Keyes leads to some great scenes and one of film’s greatest endings.
Nora Inu (Stray Dog) / Akira Kurosawa (1949). These two great Japanese actors appeared together in several movies but I don’t think they played closer friends than here where Takeshi Shimura is the wise and supportive veteran cop mentor to new detective Toshiro Mifune.
The Third Man / Carol Reed (1949). Holly Martins goes to Vienna at the invitation of his old friend Harry Lime only to (eventually) find him much changed.

Someone already added of Mice and Men, so I’ll add a few that are somewhat obscure, and possibly unknown to many even here. The Barefoot Boy, 1938. This one features über cutie, Marcia Mae Jones, in one her feistier roles, and also Jackie Moran; this would be a common pairing over the next few years. Reg'lar Fellers (1941). This “Little Rascals-esque” feature, included, of all people, Our Gang actor Carl “Alfalfa” Switzer. It was based on the comic strip of the same name.

Another animated film that is not "just a kids movie" UPIt opens with a wonderful friendship between a little boy and a little girl and evolves into a lifelong friendship between a husband and wife and then finishes up with a friendship between a grandfather-type and a lonely little boy. There is something special about this film. (The gang-dogs aside, but even they are ok.)

... Thunderbolt and Lightfoot (Michael Cimino, 1974), co-starring Clint Eastwood and Jeff Bridges. Some observers have analogized the film to Midnight Cowboy, a point raised by interviewer Brian Linehan to Eastwood in the 21:05-21:20 portion of this interview from 1974:
And although Eastwood's following film, his self-directed The Eiger Sanction (1975), is not a movie about friendship per se, it does feature a friendship (between Eastwood's character and George Kennedy's) that becomes troubled, ending on a highly ambiguous note.
Perhaps the most remarkable exploration of friendship in an Eastwood film comes in his self-directed Bronco Billy (1980), where Eastwood's protagonist goes to certain humiliating lengths to salvage a young friend (played by Sam Bottoms) who had landed in trouble for a controversial and illegal act. Aside from Thunderbolt and Lightfoot, though, the Eastwood film that leans most heavily on the concept of friendship is Million Dollar Baby (2004), where Eastwood and Morgan Freeman play aging friends. Incidentally, earlier this month, the New York Times named that movie the third-best of the twenty-first century so far (for what that is worth). Eastwood's Jersey Boys (2014), meanwhile, explores the tenuous nature of relationships—perhaps friendships—in a musical group.
Another recent film about friendship is A Walk in the Woods (Ken Kwapis, 2015), co-starring Robert Redford and Nick Nolte. Or try a film from fifty years ago: director Howard Hawks' Western El Dorado (1967, in terms of the US release), co-starring John Wayne and Robert Mitchum. In that movie, friendship trumps economic self-interest.

Citizen Kane Friendships sometimes get derailed. Kane and Leland as a prime example.
Forever Friendships : Stanley and Oliver. They may whop each other with this and that and get majorly exasperated but they are FOREVER Friends.
Shrek and Donkey fit the bill here, as well.

These were before my time, but I believe I saw them on tv when I was a kid.

spiderwort" These were before my time, " That's the beauty of films. NOTHING is before anyone's time any more. Today with the internet even more so than when we had to rely on what was being shown on the tv. Same goes for long departed singers preserved on vinyl. I used to watch the "Farmer Grey" cartoons on early B&W tv and had no idea that I was watching silent movies. Imagine my surprise and delight when I found about them in a book.

Keeping Up With the Joneses. ordinary suburban couple make friends with super spies. My favourite line is mrs Jones: "i can't help having perfect bone structure and flawless skin. I may not have to moisturise, but i still have feelings."
