It's referenced directly a couple times in the trailer, but why does Thanos only want to destroy half of the universe?? Is this something from the comics?

Yes it's from the comics (which I haven't read...I did read Thanos Annual #1 though...) At the start of The Infinity Gauntlet, the alien nihilist Thanos has collected the six infinity gems and attached them to his gauntlet. With their combined power, he becomes like a god and sets out to win the affection of Mistress Death, the living embodiment of death in the Marvel Universe. When Thanos uses his powers to kill half of the living beings in the universe, Adam Warlock leads Earth's remaining heroes against him. When the Infinity Gauntlet is stolen by Thanos' granddaughter Nebula, he aids the remaining heroes in defeating her. Warlock ultimately obtains the Infinity Gauntlet and uses its power to undo the death and destruction caused by Thanos. en.wikipedia.org/wiki/The_Infinity_GauntletEvil people always have some kind of twisted logic, and in his mind half is logical...at this somewhat arbitrary point in time. edit: OK I just saw in that wiki that this occurs after the number of living things in the universe becomes greater than the number who have died. Not that arbitrary... Don't forget that in GotG 2's stinger, Ayesha mentions/introduces 'Adam,' the next step in the Sovereign's evolution.

In the Infinity Gauntlet comic book series, Thanos wants to destroy half of the universe to win the love of Death. In the post-credits scene of The Avengers, the Other tells Thanos that the only way to destroy the human race is to "court Death". I don't know if Avengers: Infinity War will still keep that plot point.

Mistress Death thinks there are more people being born than dying so she gets Thanos to kill half the universe. He decides to use the Infinity Gems to do so.

I don't know if Avengers: Infinity War will still keep that plot point. No, he doesn't. He doesn't say the only way to destroy them is to court Death. His exact words are, "Humans. They are not the cowering wretches we were promised. They stand. They are unruly, and therefore cannot be ruled. To challenge them is to court death." In other words, challenging humanity is foolish and suicidal.

In order to "court Death", I assume the Other meant that Death must be wooed romantically.

Okay...if Marvel goes with 'courting death' as 'Wooing the Lady Death', I wonder several things. WHO is playing Death? I doubt it's gonna be Cate Blanchet, though that would have been awesome. Will Death be someone like Thanos, some super-powerful alien? Or will she be more like a being time forgot, where she's actually older than our galaxy and witnessed the expanse of universes? And also, why would Thanos think he would want to win her over - or will they just sort of gloss over that, cause he's a dude of course the impossible woman is the one he wants? Will she be the villain in A4, rather than Thanos, since there has been all this build-up for Thanos, it'd be interesting to find out he was more of an opening salvo?
If they go with (and I find this more likely, in the wake of them using, then killing off, The Goddess of Death in RAGNAROK) the idea that "to court death" is more, 'You dumb bagagaloop, these humans ain't worth the trouble trying to conquer them and their tiny planet', then he is going to be very cranky when he shows up at T'Challa has that force-field shell over Wakanda with all these people inside defending it so bravely and so well. Iron Man running around with the Iron Legion, Rhodey, and Pepper in her suit. Captain America/Steve able to hold back Thanos's punch while Thanos is wearing the Gauntlet, plus bringing his butt-kicking friends with him. Thor is running around with the last of his people, refusing to die. Loki is tricking him, to save Thor. The Guardians of the Galaxy are gallivanting with at least one of his daughters, thwarting his efforts. Wanda is using her mutant-fused-magic to protect Vision (and the Stone in his head) from Thanos and his minions.
