I haven't read too much about this movie, so I'm basing this solely off the trailer and some of those bts clips of filming.
Many people are saying that the sad-looking boy who Arthur/Joker is forcing to smile is a young Bruce Wayne. In some of the bts clips it's apparent that Joker is whipping up anti-Thomas Wayne furore in Gotham. We see from the trailer that Joker has a close relationship with his mother (she might even be his only friend). We also see that he visits Arkham State Hospital.
So, my theory: Arthur/Joker's mom ends up in Arkham State Hospital, and dies there. Arthur is angry, and believes that poor care at the state-run hospital is to blame for her death. He turns his anger on Thomas Wayne who, I believe, will be some kind of official, a mayor or governor. (Why else would Thomas Wayne be giving a news conference about a criminal?) Arthur, whose mind was already slipping, fully becomes the Joker, and in this persona he creates chaos and whips the city into an anti-Wayne frenzy. A crazed citizen (not Joker himself, though; I don't think they'll repeat that) will kill Thomas and Martha Wayne. That's why young Bruce looks so sad, and why Joker will feel a weird bond with him: Arthur lost his mom because of the government's (i.e. Thomas Wayne's) poor management of the state hospital; Bruce lost his parents because of the mob that Arthur unleashed.

I think Bruce is the dude who clotheslines Joker with the sign.

Thomas Wayne is supposed to be a more villainous-than-usual character in this film I believe. Perhaps Joker's "movement" leads to Joe Chill to killing him and orphaning Bruce?

I remember hearing a rumor at some point last year that suggested that Bruce and Arthur would be half-brothers. I don’t know if that’s true, but while I initially thought that idea was stupid, it probably wouldn’t bother me if it ends up being the case. This is basically an Elseworld story that’s presumably not getting any sequels, so they can do whatever the hell they want as far as I’m concerned.

Bruce is that little kid Joker sticks his thumbs in his mouth through the gate. I'd bet anything. Creepy!

This movie is set in the 80’s and Joker is already in his mid-to-late 40’s. The point of this theory is that he’ll grow up to be George Clooney-era Batman and Joker will be in his 60’s?
