A couple of examples I can think of:
Ben Kingsley in Iron Man 3, I really liked his character at first, I thought there was gonna be a crazy showdown with Iron Man, really pissed off when he was revealed to be a joke character.
Muldoon in Jurassic Park, he was built up as this bad ass dinosaur hunter, rather dissapointing when he was quickly killed off by the raptors.

John Cleese as Nearly Headless Nick in Harry Potter. They should have brought him back for part 5 to have that sad conversation with Harry about what it really means to be a ghost

I'd say Darth Vader in ROTJ was misused. Making him a slave of the Emperor instead of a rival took the zip out of him.
Eric Roberts and Mickey Rourke in The Expendables
ER should have escaped the island and went after the girlfriend as a hostage-then Mickey Rourke and Dolph Lundgren re-appear to help save the day.

I wouldn’t say ruined but I didn’t like the treatment.
Cyclops in the X Men trilogy. Mandarin in Iron Man 3. Scrappy Doo in Scooby Doo. Scrappy would NEVER consider harming his beloved Uncle Scooby. The ‘87 Turtles in Turtles Forever. Treated as spineless jokes to make the other versions look better. Pa Kent in Man of Steel. Mary Jane in SpiderMan 3. Aunt May in the MCU. Guenevere in Sword of Lancelot. She would never consider killing off Arthur.
At the time I was saddened by the idea that the original Star Trek Universe no longer existed because of the events in STAR TREK 09 but PICARD portrays that the timeline still exists.

I liked the relationship between Vader and Moff Tarkin. For the most part mutual respect, but I always got the feeling Moff rather disliked him and wasn't afraid to tell him to his face: "Your sad devotion to that ancient religion has not helped you conjure up the stolen data tapes". Anyone else Vader would have choked to death.

Was watching this last night. Still like it but Pa Kent’s demise in the 1978 film works better at it’s something Clark couldn’t save him from even if he wanted to. Unless he somehow figures out to turn back time.

I like that idea too. That Superman learns he cannot save everyone. Learns that lesson by his father dying of cancer or heart attack. Heck, modernize it by letting Martha die of breast cancer and his dad lives.
