Well first will their be one. I don't see how they don't put one in there so they can have Sinister 6. We have a lot to choose from. So far we have:
Spiderman 1 - Green Goblin
Spiderman 2 - Doc Ock
SPiderman 3 - Sandman
The Amazing Spiderman 1 - Lizard
The Amazing Spiderman 2 - Electro
Who do we have left
Spiderman 3- Green Goblin 2 (no chance Sony/Disney puts Franco in a Family Movie)
Spiderman 3 - Venom
The Amazing Spiderman 2 - Green Goblin
The Amazing Spiderman 2 - Rhino
Homecoming - Vulture
Homecoming - Shocker
Far from Home - Mysterio (This could help Spidie after the spell is reversed and people see that "Mysterio" didn't die so Spiderman didn't Kill him. )
Venom - Venom/Lethal Protector (Maybe you could merge the Hive Knowledge of Spiderman 3's Venom with new Venom and create more Hate for him. Venom did tell Eddie in Venom 2 that there was a hive memory that crossed universes I think)
Also could Doc Strange not be Doc Strange. I wouldn't put it past MCU/Feige to use Mephisto after all the fucking Mephisto talk for Wandavision in another project. Maybe that's why all the villains invading are ones killed by Spidie. Maybe Mephisto is bringing them in from a Hell Dimension not multiple dimensions. Or Strange could be a Skrull (Super Skrull). Strange is just acting a bid off in the trailers.

If I had to guess, I don’t think they’d have two Goblins, which eliminates both Harry Osborn Goblins. And I don’t think they’d include Venom, in any form, because then they’d have to tie him into the Venom movies or have two Venoms, and whatever’s up with the rights situation… I just don’t they’ll go that route (watch I’m completely wrong).
That leaves Rhino, Vulture, Shocker (whom I don’t actually remember), and Mysterio. Has Mephisto ever been a member of the Sinister Six? Most of my Spidey knowledge comes from the ’90s TV show and the movies, so there may be a lot I don’t know. And the most traditional members of the gang are, who, Vulture and Mysterio? Interesting that they’re the two villains from the Holland series. I found Keaton the most interesting MCU villain of them all, so I’m rooting for him to be in the movie, but that’s performance, not character. (I should love Mysterio, he’s exactly the kind of crazy comic-book character that appeals to me, but I found Far From Home’s depiction of him terrible, just like everything else in that movie.)

I remember a rumor that Rhino will at least be name-dropped. Vulture is still around, thanks to the Morbius trailer. I think they should use Mac Gargan since he appeared in Homecoming and have him become Scorpion.

I know he's in universe now and even the orange glow that flashed means it was Strange's Spell that did it, but I don't see them using Venom as one of the Villains. They've spent 2 movies trying to make him more of a Hero than Anti-Hero/Villain I don't think they will take the step back in this movie toward villain. Especially given with this many characters they won't have time to explain him and his more sinister alignment.

We can go with the shitty 80's sitcom trope and have Doc Strange be Mysterio and the whole movie + the end of Far From Home was all a mind fuck. Peter is in a long play illusion from him. lol. AKA it's been all a dream since right before the final battle with Mysterio in Far from Home.

Haven’t they already introduced Mac Gargan (Scorpion)? Wasn't he in the post credit scene of Homecoming with Vulture? If that's my memory is kinda failing me here, then yeah but not exactly on the level of having him with the other 5 established villains we know about in the trailer.

The trailer strongly suggests that Ock (maybe even Goblin too) will be fighting alongside the Spideys. So I think it’s just the five we know.

Mysterio is my favorite Spidey villain (in the comics) and I (mostly) hated the movie version. Man, would I have loved to have seen what Raimi could have done with that character! FFH was a dreadful Spiderman movie but the movie version of Mysterio was the only best thing about that shit sequel. They should have had Homecoming and NWH as the only MCU Spiderman movies. They got the look of Mysterio right, and some of his illusion sequences were well done as far as the comic booky visuals go but the tone of the movie was so off that even that stuff felt soul-less and forced. Watching that one illusion montage out of context makes it seem pretty good, but the movie had no style or personality outside of it, so it felt random as if copy/pasted from some other (better) movie. And the twist, aside from being obvious, was handled so poorly- that painful scene where he explains his motivations to the audience via a recap to his cronies? So awful.
