By "Should" I mean, do you think it serves the story of the saga better if he does know that Anakin became Darth Vader?
This harkens back to a brief exchange early in A New Hope.
Aunt Beru: Luke's just not a farmer, Owen. He has too much of his father in him. Uncle Owen: That's what I'm afraid of.
Naturally, that could be read in two ways. 1) That he is afraid Luke's adventurous spirit will get him killed like his father supposedly did, or 2) he is going to fall to the dark side like his father did.
As of now, I do not believe there is any material(Disney or Legends) that states, or even implies, that he knows the truth. So as a matter of "fact", the answer seems to be "no". However in your personal opinion, do you that the story works better if he does know?

I'm not sure he necessarily has to know he became Vader but I think from that scene it's kind of implied he knows Anakin was going down the wrong path. Apparently Owen Lars was originally a Jedi as well:- starwars.fandom.com/wiki/Owen_Lars#:~:text=In%20the%20second%20draft%2C%20Larsand - personal canon-wise - that's how I prefer to watch the OT. i.e. Owen Lars as a guy who was once a brother in arms (if not necessarily literally) with Anakin, who thought they were getting in too deep or something and decided to take up a simple life and hide out as best he could. Maybe he has a certain guilt re Anakin and that's why he tries to keep Luke away from his path as much as he can.

Did Lucas know at that point that Vader was Luke’s father? I’m not sure that was actually the plan until later. I think the line was originally supposed to mean that Owen didn’t want Luke to run off and become a Jedi with Obi Wan because that led to Anakin’s death at the hands of Vader. Which still works assuming Obi Wan told Owen the same lie he told Luke.
