Parents/children, sibling, aunts/uncles, nieces/nephews, etc. They can play relatives in the films or not.
These come immediately to mind:
Guess Who's Coming to Dinner - Katherine Hepburn plays mother to her niece, Katherine Houghton Ricki and the Flash - Meryl Streep plays mother to her daughter, Mamie Gummer On Golden Pond - Henry Fonda plays father to his daughter Jane Fonda The Long Riders - brothers Dennis and Randy Quaid play brothers in the film The Quiet Man - brothers Barry Fitzgerald and Arthur Shield play unrelated characters in the film
There are probably many more.

The Long Riders Not only the Quaids as The Millers but also David, Keith and Robert Carradine as the Younger Brothers James and Stacy Keach as Frank and Jesse James Christopher and Nicholas Guest as Charlie and Bob Ford Pretty great casting gimmick if you ask me, not that anyone did !

53-year-old John Wayne gave his 4-year-old daughter, Aissa Wayne, a speaking part in “The Alamo” (1960). While it is remarkable that she could learn and recite lines at that age, she remains the most sickly sweet child actress in movie history.

The Quiet Man is another film just chock-a-block filled with relatives : In addition to Fitzgerald and Shield Director John Ford's brother Francis Ford plays Dan Tobin - the "dying" old man Ken Curtis is John Ford's son-in-law Four of John Wayne's children appear at the horse race - including Patrick. Maureen O'Hara has two brothers in the film : James O'Hara as Father Paul Charles B FitzSimons as Hugh Forbes There's probably more, seems nepotism was running rampant on The Quiet Man set.

Cammie King Conlon I thought that she HAD to be someone's relative, but no.
"The part of "Bonnie Blue Butler" in Gone with the Wind (1939) was to have been played by Cammie's older sister; however, by the time the scenes were to have been filmed, her sister looked too old for the part. Cammie stepped in to take her place." Then she peaked at age five and retired.

Four Daughters and Daughters courageous, Four Wives, Four Mothers
Priscilla Lane, Lola Lane and Rosemary Lane played three of the Four Daughters . The fourth sister Leota Lane was not considered suitable and was replaced by Gail Page.
The Falcon’s Brother- George Sanders played the Falcon for three features, then his real- life brother , Tom Conway , came on board as the Falcon’s brother and took over the series. Conway was startlingly like George Sanders.
The Bowery Boys- since Dead End, members of the Gorcey family had been sliding in and out of the series of films built around the gang headed by Leo Gorcey. Finally, his father Bernard joined the cast as Leo Dumbroski , owner of the soda shop where the Bowery Boys hung out. Bernard stood 4’ 10’’ .

Ginger Rogers & Lela E. Rogers During the production of "The Major and the Minor" (1942), the casting department was having trouble finding an actress to play " Mrs. Applegate", the mother of " Susan Kathleen Applegate", played by Ginger Rogers... At the last minute, it was suggested that Ginger's mother, Lela E. Rogers, play the part. The production team asked her, and she accepted. It was the only film that Lela E. Joely Richardson playes Queen Elizabeth I in Anonymous and her mother, Vanessa Redgrave, played the ageing queen.
Phyllida Law has stared in several movies with her daughter Emma Thompson, Much Ado About Nothing (1993) is one.

This weekend I saw Rambling Rose (1991) for the first time. I found this movie very entertaining and easy to love. Here, Laura Dern was nominated for Best Actress in a Leading Role while her real-life mother, Diane Ladd, was nominated for Best Actress in a Best Actress in a Supporting Role. They were both great!!
I think Laura Dern also played in a movie opposite her father, Bruce Dern.
The Family Way (1966) is one my favorite comedies. While John Mills was never shy about acting with his two daughters, he was great here, playing the father-in-law of his newly-wed real daughter, Hayley Mills. The two also played opposing roles in The Chalk Garden (1964).
In the Jame Ivory film, The White Countess (2005), we have three related women in key roles--Lynn Redgrave and her sister, Vanessa Redgrave played sisters in the movie and Vanessa's daughter, Natasha Richardson played her daughter in the movie.
In Georgy Girl (1966), Lynn Redgrave's mother, Rachel Kempson also played her mother--I think that was the relationship in the movie. In any case, both were in the movie.
In The Fabulous Baker Boys (1989), real-life brothers, Jeff and Beau Bridges, played as the Baker brothers.
In Sylvia (2003), Gwyneth Paltrow played Sylvia Plath and Platrow's real mother, Blythe Danner, played her mother.
Brothers John and Lionel Barrymore both had roles in Grand Hotel (1932) and the two brothers were joined by their sister, Ethel in Rasputin and the Empress (1932).
In The Proud Rebel (1958) Allan Ladd and his real life son, David played father and son.
Brothers, Ben and Casey Affleck, appeared in the movie 200 Cigarettes (1999). I know nothing about his movie

Sofia Coppola appears in all three Godfather films.
Omar Sharif's son plays Zhivago as a boy.

Martin Sheen and son Charlie appeared together in Apocalypse Now (no idea where Charlie is in that one), Cadence, Wall Street, and Hot Shots Part Deux.
Charlie and brother Emilio appeared together in Young Guns.

Kempson played the sickly wife of James Mason .she dies early in the film and I’m not sure she has much screen time.

Angela Lansbury ‘s mother, Moyna McGill, played her mum in Picture Of Dorian Gray. She was a fine stage actress and played a lot of dress extras.

First thought was Grand Hotel with the Barrymore brothers, which is already mentioned. A whole bunch of relatives appeared in the Godfathers movies, or many Francis Ford Coppola movies. Ben and Casey Affleck in Good Will Hunting Just last year, James and Dave Franco in The Disaster Artist Will Smith and his son Jaden in The Pursuit of Happyness Tom Hanks and his wife Rita Wilson in Sleepless in Seattle Madonna and then husband Sean Penn in Shanghai Surprise Martin and Charlie Sheen in Wall Street

DAVID Ladd played a bit in Shane with his Father Alan Ladd.
Kevin Corcoran and his brothers played three of the no neck monsters of Gooper and Mae in Cat on a Hot Tin Roof. The Corcoran were in a lot of 50’s film and tv especially for Disney.
Has anyone mentioned Liza Minelli’s Appearance with mom Judy Garland in In the Good Old Summertime?
