Peter Sellers played the president, the RAF liaison officer, and the title character in Dr Strangelove. He was also supposed to play Major Kong but the role went to Slim Pickens instead.

Donald Sutherland in CASTLE OF THE LIVING DEAD played at least 2 roles ( a soldier and a woman who got a really bad early Botox treatment from Christopher Lee).

One of Karloff's finest: The Black Room
Uncouth, violent and evil twin Gregor, and his refined, goody-two-shoes brother Anton. It could be said that he actually gives three performances, with his best moments coming when Gregor is impersonating Anton. We, the viewers, know about the impersonation, and Karloff plays those scenes with the multiple layers readily apparent.
